Die Parkinson-Krankheit, die durch die Degeneration dopaminerger Neuronen in der Substantia nigra pars compacta (SNpc) gekennzeichnet ist, ist nach der Alzheimer-Krankheit die zweithäufigste neurodegenerative Erkrankung, von der etwa 1 % der Bevölkerung im Alter von etwa 65 Jahren betroffen ist. Die Krankheit ist klinisch durch Ruhetremor, Steifheit, Bradykinesie und Haltungsinstabilität gekennzeichnet. Pioglitazon, ein Peroxisom-Proliferator-aktivierter Rezeptor-Agonist, vermindert nachweislich den Verlust dopaminerger Zellen im MPTP-Tiermodell der Parkinson-Krankheit, eine Wirkung, die auf seine entzündungshemmenden Eigenschaften zurückzuführen ist. In der vorliegenden Studie wurden die motorischen und kognitiven Wirkungen von Pioglitazon, einem Peroxisom-Proliferator-aktivierten Rezeptor-Gamma-Agonisten, in einem intranigralen MPTP-induzierten Parkinson-Modell untersucht. Die Ergebnisse zeigten, dass die akute Behandlung einen gewissen Grad an Neuroprotektion bewirkte. Im Gegensatz dazu führte die chronische Behandlung zu einer Verringerung der durch MPTP verursachten striatalen DA. Diese Ergebnisse deuten darauf hin, dass die akute Verabreichung von Pioglitazon (30 mg/kg) wirksamere positive Auswirkungen auf das motorische Verhalten und den striatalen DA-Spiegel hatte.
